Heart and neural crest derivatives expressed 1 (HAND1) is a basic helix-loop-helix (bHLH) transcription factor belonging to the twist family. Twist-family transcription factors play essential roles in a number of developmental programs and function by dimerising with other bHLH members and binding to cis- regulatory elements, called E-boxes. HAND1 can form homodimers as well as interact with other tissue restricted bHLH proteins, such as HAND2. Dimer partner choice infers differential regulation on twist family proteins and is regulated by phosphorylation. Protein Kinase A and C (PKA and PKC) can phosphorylate HAND1, while β56δ-containing protein phosphatase 2A (PP2A) can dephosphorylate HAND1.
HAND1 is required for proper cardiac morphogenesis and is essential for extra-embryonic and trophoblast cell differentiation. Hand1 is expressed in several foetal tissues and in adult heart tissue where it may have a role in preventing hypertrophy.
