Sphingolipids are essential lipids in eukaryotic cells. The sphingosine 1-phosphate receptor 1 (S1PR1) mediates several autoimmune conditions, including asthma and multiple sclerosis (MS). Therefore, sphingolipid derivatives are a target to block the activation of G-protein coupled receptor S1PR1 and suppress autoimmune conditions.

S1PR1 plays an important role in cell migration, probably via its role in the reorganisation of the actin cytoskeleton and the formation of lamellipodia in response to stimuli that increase the activity of the sphingosine kinase SPHK1. S1PR1 activation is also required for endothelial cell chemotaxis and PDGF-induced cell motility.

S1PR1 has complex and vital roles required for normal embryonic heart development and cardiac morphogenesis. S1PR1 also plays an important role in regulating sprouting angiogenesis and vascular maturation. S1PR1 inhibits sprouting angiogenesis to prevent excessive sprouting during blood vessel development.

S1PR1 is a regulator of bone mineralisation and bone homeostasis.
