Nocturnin 2′-phosphatase but was believed to be a deadenylase. Nocturnin is part of the CCR4 family of endonuclease–exonuclease–phosphatase (EEP) enzymes, catalysing the conversion of NADP+ to NAD+ and NADPH to NADH. NOCT has a well-conserved C-terminal EEP domain but lacks a canonical RNA-binding domain and does not contain the leucine-rich repeat domain. It is present in two forms, depending on which initiation codon is used for translation initiation. One form results in the expression of a mitochondrial targeting signal, and nocturnin is transported into the mitochondria. Alternatively, this signal is lacking, and the protein remains in the cytoplasm.

Nocturnin is circadian-regulated with peek expression in the early night hours. It coordinates metabolism that promotes adipogenesis during the circadian clock. Mitochondrial targeting of nocturnin appears to link metabolism and the circadian clock. Deletion in mice leads to resistance to high-fat diet-induced obesity and hepatic steatosis. Nocturnin regulates intracellular NADP(H) levels and the oxidative stress response.
