Activated MAP3K5/ASK1 initiates the phosphorylation and activation of several MAP kinase kinases such as MAP2K4/SEK1, MAP2K3/MKK3, MAP2K6/MKK6 and MAP2K7/MKK7, followed by p38 MAPKs and c-jun N-terminal kinases (JNKs), which control the transcription factor activator protein-1 (AP-1).

MAP3K5 can be activated in response to stress signals, ROS; such as H2O2, tumour necrosis factor-α (TNFα) or reduced serum levels. It has been shown that a molecular target of reactive oxygen species (ROS),Thioredoxin (TXN/Trx), is an inhibitor of MAP3K5.

MAP3K5 plays a role in diseases in which reactive oxygen species (ROS) and/or endoplasmic reticulum stress are causative factors, such as type 2 diabetes, neurodegenerative disorders, cardiovascular diseases, inflammatory diseases, and chronic inflammation-induced carcinogenesis. MAP3K5 may prevent stress-induced disorders and protect from bacterial and viral infection.
