ANO1 is a Ca2+-activated Cl− channel protein, expressed in the epithelia, with this expression in the Interstitial Cells of Cajal (ICC) being responsible for the slow wave peristalsis in the gastrointestinal tract. The ICC are the pacemaker cells for the gut, involved in the propagation of the slow gastrointestinal wave, required to move ingested material through the gut. Blocking or knockout of ANO1 is reported to result in the loss of organised smooth muscle contraction responsible for peristalsis, suggesting ANO1 is essential for the normal function of gastrointestinal physiology. However, ANO1 is found in all ICC cells, including those not responsible for pacemaker function, implying there are unknown additional functions.
In addition to the function within the gut, ANO1 is expressed in several epithelia, controlling several cell functions including proliferation, survival, migration and secretion. ANO1 was initially discovered due to high expression of the channel proteins in gastrointestinal stromal tumours (GISTs), and was previously known as DOG1 (discovered on GISTs).
